The actual clearance is issued when you call them, (radio at towered field, land line at uncontrolled field) field and put your clearance “on request”.

Takeoff clearance is given by the control tower. You simply line up at the departure end of the runway, call the tower, and tell them you’re ready for takeoff. Normally, if there are no aircraft ahead of you and no conflicting traffic, you will be cleared for immediate takeoff.

When can ATC not issue a takeoff clearance immediately?

This sentence is used only when ATC can not issue a takeoff clearance immediately (e.g. because there is still traffic on the runway). Controllers must not use this command unless needed.

Who is in charge of ATC in aviation?

That being said, ATC exists solely for YOU as the pilot, and as pilot in command YOU are in charge of the safety of the flight. A routine example of this is if ATC clears you for takeoff and says “no delay, Learjet on 2 mile final”. You don’t have to accept this takeoff clearance.

When does a pilot have to tell ATC when catching the localizer?

What clearance does the controller give the pilot to take off?

The controller authorizes the aircraft to take off. This is the one and only clearance that gives a pilot permission to take off. If you are holding short of a runway, this clearance also gives you permission to enter it and align. The controller does not need to tell you to line up and wait beforehand.
